Here Rich posted his guide to yeast slanting. In the last part of the guide he talks about growing yeast for pitching. This may help some.

What I have been doing is making a sterile starter solution in the same test tubes as slants fill about 1/2 full, I make a whole bunch at 1 time. then I inoculate them from the slants, after 2 or so days I have a pitchable amount for 500ml, and then build it up from there.
