I mostly do smaller starters, but I bought a 1000ml Polymethylpentene Cylinder ( http://www.usplastic.com/catalog/item.a ... &catid=649 ). I pour the slurry into the cylinder, add 4x water, shake it up, let it come to the top and then pour the desired amount into an erlenmeyer flask.

I store the yeast from a previous batch in a 1 gallon glass jar until I'm ready to pitch. I'll decant the beer off the top until it's about 50-50 beer and yeast. I'll stir up the yeast and beer with a sanitized spatula.

The pitching rate calculator is providing just the yeast slurry volume, so I double the amount since my solution is 50% yeast slurry and 50% beer. Then I pour yeast-beer solution into a 500 ml beaker to the 2X amount.

I measure out my yeast with a few glass beakers similar to:

I mainly use a 400ml beaker. I keep them clean, then soak them in my bucket of star san, and then let them dry upside down on a clean sheet of aluminum foil. After they dry, they are ready to measure out the saved yeast.
